全自動外泌體熒光檢測分析系統ExoView是美國NanoView Biosciences于2018年研究推出的,該系統一經推出,便引起了外泌體領域科研工作者的廣泛關注。僅兩年在全球已有100多個實驗室采用該技術,發表文獻近百篇,刊載在Cell, Cell Biology, Journal of Extracellular Vesicles等期刊。ExoView的表征,為科學家提供了一種無須純化即可直接對血漿、血清、尿液、培養基中的外泌體進行表征分析的新方法。能夠提供單個外泌體水平下的全方位外泌體表征信息(包括粒徑大小、分布、計數、攜帶蛋白表達與生物標志物(CD9,CD81,CD63等)共定位等),以及多層次和全方面的外泌體測量解決方案。能夠幫助科學家更深入地了解外泌體與疾病之間的關系,助力疾病診斷和新藥開發。
ExoView? R200全自動外泌體熒光檢測分析系統優勢包括:
? 特異性捕獲:芯片上可包被多達6 種捕獲抗體,特異性捕獲含特定蛋白標記物的外泌體。
? 絕對陽性外泌體計數:芯片捕獲外泌體后,可通過單粒子干涉反射成像傳感器(SP-IRIS)技術直接檢測樣品中外泌體的數量。
? 單個外泌體蛋白共定位分析:檢測每個外泌體的熒光信號并進行統計,可獲得熒光共定位信息,用于分析樣品中不同表型外泌體的比例。
? 無需純化:使用抗體捕獲模式,防止樣品中雜質影響結果,可直接檢測血液、尿液和細胞培養液中的外泌體,未純化樣品的測量結果與純化后基本一致。
? 粒徑分辨率高:高精度SP-IRIS技術,可檢測≥50 nm的外泌體,測量結果與電子顯微鏡檢測結果基本一致,并統計生成外泌體的粒徑分布結果。
? 可檢測外泌體內容物:試劑盒配套相應的穿膜劑,可穿透外泌體并對外泌體內容物進行染色并檢測,如下圖所示,Non-permeabilized時只能檢測到跨膜蛋白CD9的熒光信號, Permeabilized后即可檢測到外泌體內容物(如PD-L1、P-tau等)的表達。
